Good evening (at least in me time-zone)
I have been having some trouble with some routine Immunological staining. What the ideal conditions for IHC slides before staining is? Should I leave these slides to drain overnight at room temperature or use the slides dryer for 3 hours at 45 degrees? By the way, What is the appropriate temperature for a tissue flotation water bath?
I hope to receive any information, suggestions, leads, comments, questions, and/or thoughts you may have.

I have dried IHC slides both ways...O/N at room temp and in a slide dryer (60C for 30 minutes) with no difference in staining. Thus, your problem with your IHC staining might be something else. Your water bath temp depends on the melting point of your paraffin and should be several degrees below that melting point...warm enough for the section to flatten out, yet not so warm as to melt the paraffin completely away. Hope this helps.
