I would like to sequence genomic DNA with the Nextera DNA sample preparation kit. For those who used it, you know for sure that it's very expensive! We were thus wondering whether it was possible to divide the number of reactions by 2, as we often do for other kits?
If anyone tried this, could you please let me know how it did end after sequencing?
Maybe! Check out this article: "A cost-effective method for high-throughput construction of illumina sequencing libraries." They basically cut the volume of their reactions and still have good results. They also make use of homemade index pcr primers that further reduces the cost. I have not had good luck using the homemade index pcr primers but I have been attempting to use them on amplicons and not on whole genomes.
We have found that we can cut the volumes in half but only for certain types of DNA samples (GC-neutral). In this case, everything should be cut in half (including input DNA) to maintain stoichiometry.
