I need to collect human granulosa cells from a fertility clinic. The clinic does not have a refrigerated centrifuge. The protocol asks for 15 min at 1000 RPM at 4°C to separate them from follicular fluid and then shorter centrifugations for lavage and RBC lysis. A few colleagues told me that I could centrifugate the cells at room temperature, but in less time. If I do that, how much time should I centrifuge them, and at how many RPM? The objective is to separate the cells for qPCR and western blot (not for culture). Thank you so much!
