That depends on a number of factors. 1) the proteins you want to detect must be of different sizes so they can be well resolved on SDS gels, 2) the antibodies don't cross-react with proteins of interest, and 3) the antibodies are well characterized with no overlapping background bands.

Prior to doing the multiplex, try doing singleplex westerns with each antibody to ensure the antibodies do not cross-react.
