I have expressed a human membrane protein complex in insect cells (Sf21 cells) using MultiBac expression system. At time of expression the insect cells appeared to be old- but were still growing normally. My question is whether the heterologous protein expression can perturb the membrane protein insertion- in a way that the changes the native structure?

I don't know much about your expression system but I have some experience with membrane protein in bacteria. Usually you several complex systems to do the integration/folding of membrane proteins. Usually when you overexpress a membrane protein you can saturate those systems perturbing the integration of all proteins in the membrane. This process include several chaperones that might affect the folding process of that and other proteins.
