I'm working on protein induction in e.coli using IPTG and for me the protein is not properly induced. I think E.coli is not having good growth. After doing overnight induction, the growth media is not so turbid. Media component is yeast extract, tryptone and glycerol. I'm using yeast extract from SD FINE CHEM company and tryptone from LOBA CHEMIE / HIMEDIA. I think the problem is in my media quality.
Different bacterial species need a favorable environment and nutrients for their growth.So if you think the used nutrient media is not good quality,you should try with another good quality media.You should take decision depending upon the growth of bacteria.If it takes to long time it is not favorable for their growth.
usually E coli gives good growth overnight, provided all growth parameters are set optimum. For example temperature ( use shaker incubator), pH and inoculum (log phase culture). In the case of media its better to use Himedia Nutrient broth instead of constituting using different components.
