I'm trying to wash the gel-filtered platelets that give me supernatant and pellet, and then store them at -80 for a day. How can I do that without causing activation of the samples or platelet pellet death?
I would be most appreciative if anyone could kindly guide me on this.
Even putting platelets in a refrigerator leads to artificial platelet activation.
There is no way to freeze platelets without influencing the activity.

DMSO (6 percent) has been effectively used for the cryopreservation of platelets over the years. I recommend preparing a highly concentrated stock, and measuring some activation markers of platelets including CD62P (ELISA kits are also available as CD62P can detach from platelet surface, therefore, measurable in plasma), VWF, or serotonin (HPLC is the best method) then adding DMSO, freezing and thawing and remeasuring to make sure that platelet activation does not occur during the process.
