I want to ask does EGM 2 basal media can be used without supplements to study HUVEC proliferation or requires the addition of the supplement.
Basal medium have no growth factors or serum and cells can not proliferate or survive without.
You need to add supplement and growth factors to induce proliferation and survival.

To study HUVEC proliferation, EGM2 basal medium without supplements is not recommended. Usually, the basal medium contains amino acids (essential and non-essential), vitamins, organic compounds, trace minerals and inorganic salts which are not sufficient to support proliferation and survival. Presence of supplements in the form of hormones, growth factors and other proteins will have a positive effect on proliferation and growth.
These cells can be passaged until P-5. After P-5, the viability and functionality may be compromised.
