HL-60 cells are cultured and expanded in IMDM medium containing 20% FBS. However after thawing I observed that these cells made disordered aggregates although they are still being proliferated and expanded! Now, I don't know exactly that they are being changed in their gene expression pattern thus being differentiated or they might have Mycoplasma contamination?
And it is affecting my results when I do cell viability assay: CCK-8 where the OD values of the control group (cells plus medium only no drug added) do not reach 0.8~1 even after 3hours of incubation.
