I am trying to use DNA FISH to detect plasmid DNA after transfection. I am curious if one should expect to see signal in the cytoplasm only or cytoplasm and nuclei? Any literature suggestion is highly appreciated.
Hi, I think it depends on your sequence , therefore you should select a unique sequence only found in plasmid , maybe you can target origin of replication site of the plasmid.
I would suggest you shoud follow this paper, hope you wil find your answer.
