Poly(ADP-ribose) polymerase-1 (PARP-1) is a nuclear protein that is involved in the DNA base excision repair system (BER), where it is activated by DNA strand nicks and breaks. Using NAD+ as a substrate, PARP-1 catalyzes the formation of polymers of ADP ribose units on several nuclear proteins including histones, transcription factors, and PARP-1 itself. However, PARP-1 has been implicated in many other biological processes. In fact, a clear definition of its function remains elusive. Given this uncertainty regarding PARP-1 function, can detection of PAR polymers in cells be considered an indicator of DNA stress, or, can it equally well be an indicator of some other molecular process?
