I am studying the effect of different fatty acids on LPS induced macrophages (J774A1 and THP-1). I use saponified fatty acids in complex with BSA, but observed that while i would like to study Fatty acids effect on inflammation, BSA control (even without the FA) addition to LPS stimulated macrophages resulted in increased TNFa secretion, as if BSA was also an agonist...And I have tried several FA:BSA ratios (5-50:1), and also several sources of BSA (Merck, Sigma) all were fraction V free fatty acids. Can anyone suggest a solution for avoiding this 'side effect' of BSA ? Thanks
