I want to measure Hsp60 in plasma samples but I am using monoclonal capture antibody with ascites fluid and diluted it in PBS but the assay did not work , and the optical density values were very low. Is any one have a solution to this problem ?
A positive control might help you to find out where the problem with the assay is occurring. If the "standard curve" dilutions are working fine, then its not really needed. However, something like a purified Hsp60 (http://www.enzolifesciences.com/ADI-SPP-540/hsp60-human-recombinant/) might be useful. By using this, you can see whether the negative results is dependent on your kit or whether it is to do with the samples. Best of luck with sorting it out
