I am doing DNA extraction from bacteriophage but still i get genomic DNA after adding of both DNase and RNase. so I was wondering is it possible to add DNase after perciptiation with PEG 8000 and before adding of proteinase K and 10% SDS?
Should be OK. You could test, determining whether pfu titer effected.
The phage DNA being encapsulated, should be protected.
Hi, I think you can not, as it might digest your phage DNA after doing phage DNA extraction as the DNase is still in your sample in active form, that is why the protocol mention that the DNase should be added before chloroform ( because the chlorophorm will deactivate the DNAse and RNase).
We usually use PEG6000 at a concentration of 8-10% for phage precipitation at an early stage of purification. This is better because PEG8000 can precipitate more chromosomal DNA that has not yet been digested with DNase.
