Thank you very much. I guess I will use FACS to determine the TCID50 then. But I have heard that FACS usually lowers the value a bit. So I am still struggling in picking up a reliable method.

Thank you very much for the info. I somehow gave up considering PCR. Instead, I am going to apply the plaque assay. I am very new in this area, so I don't know which method works better for my purpose. My study has not been initiated, I will update here once I have some data. Thanks.
