I am trying to develop orthotopic lung cancer model in nude mice. I want to inject the single cell suspension directly into lung parenchyma without surgery . After injection of cell into the lung parenchyma, how do I confirm tumor formation by physical observation?
For non-invasive methods, you usually need an imaging technique. You can luciferase your cells and monitor by bioluminescence or you can just perform MRI, CT or PET scan. The main issue for orthotopic implantation is the accidental spread through the pleura, so may want to avoid this by a good implantation technique.

You can also perform intratracheal injections. This tends to form a tumour concentrated in one region of the parenchyma (adjacent to airways) rather than tumours spread throughout the lung, like IV injections. You will need to infect your cells with luciferase, this allows you to do in vivo imaging with an IVIS spectrum or similar. Mice with lung tumours tend to cope really well with the tumours, I would say you can't tell from the mouse's behaviour if they have a lung tumour or not.
