Many methods available in many journals. Ho ever, I do not know what is the appropriate method to get good quality DNA from actinobacteria. Because DNA quality will affect the sequence quality I would end up with poor quality sequence. Hence I am expecting th best and most efficient method to get good quality DNA from marine actinobacteria.
I figure that you want to get gDNA for WGS and or MatePair sequencing. For that,
you might try the attached protocol. Works for most Corynebacterium and Streptomyces species as well as a few other Actinobacteria I used it on. I use the obtained DNA to create 8-15kb Nextera MatePair libraries, so the quality is good enough for sequencing :-)
Caveat: With Streptomyces and other filamentous growing bacteria, getting the amount of starting material right can be a bit tricky. If you have enough material, you might want to test different amounts of starting material.
