It isn't a dye, but have you considered using genetic constructs like gCAMP?
Hi Leo and Bhavin. Thanks a lot for your valuable comments. Yes I am trying to use both gCAMP constructs and the dye to get the readouts. Leo, all gCAMP lines are UAS lines, so I am thinking if it is wise to rely on those results. Bhavin, have you tried FURA-2 yourself? Or do you know anyone who has tried? I was looking for a detailed protocol on analysis.
I would say that it is quite standard to use the gCAMP lines now. They have been used primarily (and successfully) in neurons, which are a sensitive system in themselves. But you are totally right; I would use multiple methods to validate your experiment.
