Dear colleagues!

I currently work with alkaliphilic fungus from soda solonchaks (S.alkalinus). For the study I use buffered media based on TRIS-HEPES buffer solution to achieve various pH. But TRIS-HEPES solution provide pH range just about 7,5 - 9.0.

Can Anyone suggest the organic buffer which may provide wider pH range (I'm interested in pH values higher than 9: up to 11).

And one more question:

We now use malt extract that can decrease the pH values in medium when we join buffer solution and a base of the media. What else can we use as a carbon source to keep stable pH of our media?

Note that the species of interest lack invertase. So carbon source such as sucrose or other carbohydrate compounds with analogous binds cannot be consumed.

Sincerely,

Maria
