I currently use Liberase TL to digest collagen droplets, and also isolate TILs from tissues.
This product is currently not available from Sigma (the sole supplier), Liberase TM is too harsh, and Liberase DL is also not currently available.
Can anyone recommend an alternative, such as Dispase II, or a collagenase family member? Thanks in advance.
I know that Roche now sell a Liberase mix that is called T-Flex that contains the two enzyme mixes in seperate vials:
- collagenase I/II mix
- neutral protease
The differences in "harshness" between the TL and TM liberases is the collagenase/protease ration, and you can adapt it to your own specifications with the Liberase T-Flex.
Liberase TL has a collagenase/protease ratio of about 40-50 (U/U), so it is easy to achieve this ratio by mixing the collagenase/protease from the T-Flex kit

Unfortunately this product does not seem to be distributed in the UK.
Usually Roche distribute via Sigma, and they do not carry this product.
Hello Iain and Russell, we are having the same problem with Liberase TL.
It seems also that in Spain Roche is not distributing Liberase T-Flex. However, I have found this other company "Creative Enzymes". It seems that it is exactly the same product. Do you have any experience with it. Thanks a lot.

Have you tried Accumax from ICT, Inc. It works well for dissociation of mouse tissue.
