I am trying solvent extraction of extracellular compounds using different solvents based on polarity. I find the compounds are miscible with almost 4 of 5 of the solvents used. Is there any recommended gradient of solvent for extraction. My culture is streptomyces.
Hexane, chloroform, ethyl acetate, methanol would be good for you to extract the low polar compound to high polar compound. Can you mention the exact protocol you are following?
I have tried all solvents what ever you have mentioned. But i need a graident solvent which can pull all the active compounds... Suggestions please??
have you never used a solvent mixture?Do you extract a solid or a liquid culture? If your sample is solid the most useful are hexane /acetone or metanol/methylene chloride. If it's liquid you must control the pH of your solution: if you have acidic compounds you can extract very efficiently with ethyl acetate after acidification and you will have both polar and non polar compounds in your solvent. The same if basic.

Hai Chiaro thanks for the suggestion..My compound is a extracellular metabolite released into the broth by the organism. When pH was monitered it showed acidic profile. Im extracting with ethyl acetate. I feel Ethyl acetate is unable to extract all the compounds because the liquid was tested for activity as the control after complete extraction and still activity is seen.
