I am planning to perform a hydroxyproline assay on fibrotic lung tissues. We are not planning to purchase any kits and wanted to know if someone has been using their own protocol for measuring the collagen content.

I would also appreciate if one could share the preparation of reagents - especially chloramine T, oxidation buffer, and ehrlich's reagent preparation.
