I was given some HCT-116 cells and I plated them in 10 cm dishes about a week ago however there are maybe about 20 cells in two of the dishes. One of the dishes has taken off quite well and I have many cells but the other two dishes are growing so slow.
Has anyone else experienced this with these cells before? From what I understand they grow very quick. Should I trypsinize the low density ones and move them to a smaller dish do you think that would help?

HCT-116 colon cancer cells are quite easy to grow and proliferate in vitro, so that they normally take 2-3 days to become confluent in 10cm dishes.
The trouble-shooting you have to perform is as follows;
Bacterial/ Viral Contamination
STR analysis
Failure to add appropriate serum into the growth medium
These cells should grow very quickly and double within a couple of days. It may be you made an error in your cell count before plating of in the pipetting. I personally dont think you should continue with the cells that have not been growing as they will have been under stress due to low plating numbers and are obviously unhealthy. For good experiments you need happy logarithmically growing cells so i wouldnt use them. Hope this helps
Thanks for your help I have since thawed down new cells and got rid of those slower growing ones.
