I am planning to do western blot for a start to analyse CD133 expressions in my cells, and therefore are using unconjugated antibody, which I hoped can be used for other applications such as immunostaining and FACS for subsequent analysis. Miltenyi is a good option but apparently their CD133/1 have not been tested for WB so need to get another one the WB. I am trying to low-down my budget, using one ab for many applications, but at the same time I don't want to compromise my results. Has anyone any suggestion? Or has anyone uses CD133/1 for western blot with good result?
Dear Ain, Saleh forwarded you a spec-sheet for CD133 from Miltenyi which mentions IHC and the two papers using this antibody.The Promin (CD133) in the eBioscience catalogue is in the table listing antibodies validated for IHC but there is no mention of WB.
There are numerous other sources listing anti-CD133 which are validated for more than one technique: EMD Millipore (WB, IHC), Biobryt (WB, IHC-P), Fitzgerald Industries Int (WB, IHC), GeneTex (WB, FCH, IHC), MyBioSource (WB, IHC), BioLegend... and many more. You could check spec-sheets for publications and/or e-mail suppliers for help - some may even have trial size vials of the antibodies. Good luck with your project.
