In Western blot analysis of cell lysates, detection of both Janus kinase 3 (JAK3), and phosphorylated JAK3 is very difficult. We have tried several antiobodies /Cell signaling, Santa Cruz etc, but none worked. With JAK1 and pJAK1 we have no problems, so our protocols are OK.
Anti-JAK3 antibody [EPR908(2)] (ab92595) .... Have a look at this. It might be useful to you .
Maybe you can work around these problems by performing immunoprecipitation experiments (IP with JAK3 and determination of phosphorylation-status with an anti-phos antibody)?

If you don't have good results, you have to 1/ Change the percentage of the gel (between 4 and 7%). 2/ Extend the migration time.
