I got an interesting result of Western Blot analysis related to a compound that we are working on. The illustration of the Western data is attached in the link. It seems that the compound inhibits autophagic flux (Based on p62 accumulation). However, I was not able to understand why and how p62 levels are going down with increasing drug concentration, when co-treated with Bafilomycin A1. And why are Phospho 4EBP levels going up with increasing drug concentrations when co-treated with Bafilomycin A1? The compound is a lysosomotropic compound and alters late endosomes as well. Can anyone provide any suggestions?
