I am trying to transfect plasmid DNAs into the U266 cells. I tried Amaxa electroporation, Lipofectamine, and ViaFect. None of these techniques is working. Can anyone please suggest a better way to get DNA into the U266 cells?
Hi Prahlad, I also found it difficult to introduce DNA into multiple myeloma cells. I recommend using lentiviral transduction, which proved to be very efficent and straightforward. Another option you might want to consider is Streptolysin-O - a bacterial toxin which temporarity and reversibly permreabilizes the cell membrane so that DNA can get in. It goes without saying that Streptolysin-O is highly toxic to cells and requires careful titration. As Strep-O forms pores, you will also have to take efflux of cytoplasmic material into consideration. Cheers, Norbert
