I'm trying to repeat the experiment from 1977 - isolation of platelet plasma membranes on Poly-L-lysine coated Biogel beads. The authors don't mention the detail in which form was the Poly-L-lysine (HBr or HCl) so I don't know which one to use. Also, I can't get the answer from Sigma about the products that they sold at the time. In the attachment- paper with the protocol for preparing the beads.
Note, that in the paper the chloride salt forms are used for all of the other cationic reagents. My guess is that the counterion doesn't matter, but since the chloride salt form of poly-L-lysine is available I'd use that.
They used poly-L-lysine cca MW 80 000, and now I can only get poly-L-lysine- HBr (MW 70 000-150 000) and poly-L-lysine- HCl (MW 66 000).
Also, HBr gives better coating features and at first I was thinking that it must have been HCl as you mentioned but than, maybe, they used HBr and added chloride later to remove bromide.
counter ion does not matter in that kind of experiment, i think. Br salt of the polimer is better to form a precipitate mightbe. moreover, it has lower amount of polimer considering weight ratios, so it is better for the company... that sells by weight.
