If the protein is not supposed to be secreted then you would not expect to find much in the extracellular space. It would seem you just have low expression (or high turnover and degradation)
Exactly this protein is a nuclear binding protein and should remain inside the cell, but when I measured it in the serum, I found its concentration was high, therefore I was confused as, to why its expression is low inside the cell and became high in extracellular.
