if a protein has low intracellular expression levels, it is likely that the amount of protein secreted into the extracellular space will also be low?
If it is a secreted protein, the concentration inside the cell may be low because it is secreted as it is made.

But No the protein should always remain inside the cell and not be excreted out.
If the protein is not supposed to be secreted then you would not expect to find much in the extracellular space. It would seem you just have low expression (or high turnover and degradation)

Exactly this protein is a nuclear binding protein and should remain inside the cell, but when I measured it in the serum, I found its concentration was high, therefore I was confused as, to why its expression is low inside the cell and became high in extracellular.
Maybe you have a lot of cell lysis. Try checking for some other proteins in the extracellular serum

I do but this slightly higher concentration was seen in diabetes patients compared to the control group.
Its intracellular expression in previous studies of diabetes patients showed low.
