For screening ,Iodine staining or Ruthenium statining is quite effective method.

I have no idea if ready made substrate is available for estimation of alpha-glucoronidase .activity.

However, I think you can explore calcoflour binding with pectin? I wonder if it binds with pectin. It binds well with cellulose and chitin.

If the calcoflour binds with pectin, then a quantitative assay for pectinase could be developed.
