I am just planning an experiment to measure intracellular calcium in fish sperm with Fluo-3/4 AM using Flow cytometry. I have a question about the mechanism about Flow cytometry: is it possible that the flow filter receive the fluorescence signal from the extracellular solution? Is the washing step in the protocol to eliminate this effect? If I do not wash it after staining, would the extracellular signal be received?
