Sex determination in spiders using flow cytometry.
What are the mechanisms of sex determination in spiders ?
Are there sexual chromosomes like in insects, birds and mammals ?
If so there may be a difference in the Genome Size/%GC between male and female.
To measure %GC you will have to use Hoechst 33342 (%AT) or Mithramycin/ Chromomycin A3 (%GC).
Problems come with particular excitation of these fluorochromes : 365 nm for Hoechst and Mithramycin, 458 nm for Chromomycin.
