I want to use a von Kossa staining protocol to quantify cardiovascular calcifications. However, in our lab this protocol has not been developed yet. Does anyone know/has a proper working protocol?

I used this kit on P1 limbs and p1 spine for mouse (tissue cut without decalcification, easy to cut at that stage). This kit worked beautifully for my tissues. It should work for your purpose, I believe.

I have used this method for staining.
1. Wash the cells by PBS for two times
2. Cell fixation using 4% paraforlmadehyde for 30 min at room temperature
3. Wash with PBS for two times
4. Dissolve 100 mg of Silver nitrate in 2 ml water (5%) and add 500 ul to each well for 20-30 min under UV light
5. Wash with water for two times
6. Dissolve 100 mg of sodium thiosulfate in 2 ml water (5%) and add 500 ul to each well for 5 min.
7. Wash with water for two times
8. Add 500 ul of Nuclear Fast Red staining solution and keep for 5 min.
9. Wash with water for three times
10. Microscope
