Hi everyone I  have read the papers in most of the journal cytokinin and auxin use for shoot regeneration is 2-3mg/l (BAP) and 0.1-0.5mg/l (IAA or NAA). in my case I have used up to2- 8 mg/l BAP and 0.1-0.5mg/l IAA. But not getting a good response.

Do you think I have to use higher conc. of cytokinin than what I have mentioned above with or without auxin? 

Another question is when we get multiple shoots from callus how should shoots separate before placing into the rooting media. Because when I was trying to transfer my regenerated plant I didn't understand how to place into the rooting media.

you can increase cytokinins concentration in the media at a certain concentration and used (BA and kintin ) together, with or without auxins , and you note response of callus on developement and regeneration of shoots.

must you separate the shoots carefully and thoroughly, must at all shoots containing on base when it cultured on rooting media and lenght appropriate

I am also facing the same problem as calli increased in size instead of shoot formation when incoulated into MS media (NAA 0.25 mg/l, BAP 0.5 mg/l)and TDZ 0.25 mg/l.

Kindly suggest for the same.
