any one have a protocol for preparation of mevalonate from mevalonolactone can send to me because i prepared it using 2mM KOH and adjust the PH to 11.5 after incubation for 30 at the incubator.it is not work with me and repeat the experiment three times with different concentration.
As far as I could see, there are at least three problems (either simultaneously):
1. the amount of KOH was not enough; 2. pH 11.5 is not enough high; 3. at the applied temperature the hydrolysis is not enough fast.
Although, I am not familiar with the mevalonolactone stability, but first I would boil the strong basic solution for an hour or more, and I would use KOH excess (half mol?) and some ethanol (30-50% to water) also, to increase the mevanololactone solubility.
If it does not help, perhaps a two step procedure might go. Step one: using anhydrous MeOH and NaOMe (1.5 mol for the lactone) overnight or boiling. Then Step 2: dilute your solution with water and boil or leave it for a certain period in a warm place. Many times methyl esters are easier to hydrolyze than the lactones . Additonally, when you are removing the MeOH the the solution becomes more and more basic which can make the hydrolysis complete.
