I will work with HeLa cells and expose them to di-2-ethylhexylx phthalate (DEHP). I've been reading articles to choose DEHP doses to try in the MTT test. Is there any different guideline or something else to check it? Do you have any recommendations?

I have never used DEHP as a treatment, however I tend to believe that the procedure can be done as standard practice.
If for example you dont have any initial values to begin with, you could do a wide dose-response curve. E.g: start with 300 uM, and do a dilution series from this stock (250, 200, 150, 100, 50, 25, 0 uM). If the first curve is not fitted nicely, for example at 50 uM you have already a huge drop in viability that accounts for more than 80%, you can adjust the curve to those values, e.g start at 50, and dilute to 40, 30, 20, 10, 5, 2 uM.

I think as you said, and i did an experiment plan like that. I reviewed four studies in which various cell lines were exposed to various phthalate derivatives. and as a result, I determined a dose range as follows: 1-2.5-5-10-20-30-40-50-60-75 µM.
In the studies I reviewed, the concentration units were nM, µM, and mol/liter. I moved by turning the units together. Accordingly, I determined the concentration range and a common unit. Is there anything wrong with this?
