Would you mind sharing your experience regarding nano-LC systems for shotgun quantitative proteomics (MS/MS)? I would also like to explore if any particular nano-LC model that is more compatible to the Orbitrap systems.

May you please shed some light on the below questions:

1.      How important is the usage of 2D-nanoLC (over 1D-nanoLC) on shotgun quantitative proteomics (e.g. for complex mammalian samples)? Is it a must?

2.      How about the “on-line” or the “off-line” set-up for 2D nano-LC-MS/MS. Whether the "online" or the "offline" mode is more popular?

3.      Searching through a couple of literature, I came across a few popular nano-LC models: (a) Dionex UltiMate 3000 RSLCnano; (b) Thermo Scientific Easy-nLC 1000; (c) Waters nanoAcquity UPLC

May I know which model you think is more compatible to Orbitrap systems? Have you tried 1D or 2D Nano-LC-MS/MS on anyone of them?
