Hi, I am working on a project to analyze protein interactions from PBMC. Our problem is that PBMC cannot be extracted from human blood samples immediately, so we just centrifuge the samples to discard plasma and frozen the remaining part. Can anyone give me some help to extract proteins from these frozen blood cells? Thanks so much.

There is nothing special about this. Simple thaw (if not already in a pellet), spin down and add your lysis buffer of choice.

In my experience, there is no difference reagrding protein isolation prom blood samples compared to other tissues, cells etc.
Maybe, try a RIPA buffer-based isolation protocol (e.g. see Circ Res. 2005 Jun 24;96(12):1233-9.).
