Hi,

I Have a very basic knowledge on HPLC.

I am studying on a crystalline, hydrophobic/low aqueous solubility polyphenolic compound. I bought it from a supplier at >98% purity.

HPLC info:

HPLC with DAD detector

Flow rate: 1ml/min

90%Methanol:10%H20+0.1% Formic acid

Wavelength: 245nm

Injected volume: 20µL

1) Sample: Water Only

Retention time:

2.572min

2) Sample: Methanol only

Retention time:

2.418 min

3.226 min

3) Sample: Polyphenolic compound in Methanol; concentration 10ppm (10ppm to 1000ppm shows the same result); there is always a biggest peak at around 9.8++mins)

2.401

2.541

3.227

9.989 min  ------------ Peak (compound/polyphenolic retention time)

4) Sample: Polyphenolic compound in H20 (30mg/ml).... Filter with 0.45um before diluted in methanol  (1000ppm to 30000ppm shows the same result)

5.504 min (what is this?) 

* MISSING Any peak at retention of 9+*

5) Sample: Polyphenolic compound in H20 (30mg/ml).... Centrifuge 4000g.5m. Supernatant diluted in methanol (No 0.45um filter step as compare to number 4)  (1000ppm to 30000ppm shows the same result)

Retention time:

2.941 min

5.661 min (what is this?) 

9.873 min 

Anyone? HELP!!

I would like to understand:

A) what is this Extra retention time at 5.6+minutes that does not appear when the polyphenolic compound is dissolve in methanol only?

It could not be solvents peak as water and methanol does not have any peaks at retention time of 5+ minutes

B) Why does my compound polyphenolic in water when filter with 0.45um before diluting in methanol does not show its retention time at 9+minutes as oppose to result of number (5)?

c) I wanted to test the solubility of this phenolic in water/aqueous. However, if I filter it with 0.45um, I cant get any peak at "Im assuming its retention time at 9+minutes;based on Run(1)". 

I really have no idea.  :C
