I am purifying virus through ultra centrifuge. I want to make gradient using cesium chloride. (1:1, 1:2, 1:3, 1:6). I found the many protocol stating that 3g + 17ml gives 1:1 density. But i want to know the exact calculation and principle behind this density gradient preparation. so that i can able to prepare any density with any volume without any doubt or confusion.

If you take the data on concentration and density (for example at https://en.wikipedia.org/wiki/Caesium_chloride) and put it into Excel you can get an equation to fit the curve. I have attached an example spreadsheet.The equation of the regression line can be used to do the calculations:
y = -53.746x^2 + 224.26x - 169.69
y = the required density
x = concentration (%weight/weight, caesium chloride/water)
So if you want a 1.2g/ml solution you calculate the following:
y = -53.746*(1.2*1.2) + (224.26*1.2) - 169.69
I have included a calculation in the Excel file - just change the value in cell G3, press return and the numbers will be calculated automatically. For different volumes, just change the two numbers (weight of CsCl and the weight of water) in proportion.
