In one of my studies, I investigated miRNA-miRNA synergistic interactions by using bioinformatics analysis. Two miRNAs were revealed to have diametrically opposite properties of acting with other miRNAs. One is very willing to synergistically act with other miRNAs whereas another plays roles in relative isolation, namely, non-synergistic with other miRNAs. A new problem comes. How to validate the different collaborative tendency of miRNAs with other miRNAs in vitro? May be hundreds of miRNAs expressed in a cell.
First, I would confirm the expression pattern of your interested miRNAs to make sure they synergistically/non-synergistically expressed with other miRNAs(as your bioinformatic analysis). Then I will try to do overexpression (or knockdown?) of each of the two miRNAs and look at the expression of other miRNAs. Just be careful with your experimental design and let me know if this answered your questions. THX
Thanks Shuai. Here, the miRNA-miRNA synergistic interaction is referred to synergistic action by miRNAs, not expression. Synergistic action can be anti-apoptotic action, or other effects. To more accurately express my intent, I attached my article regarding miRNA synergy prediction as background information for this question. My bioinformatics analysis revealed huge difference of miRNAs in the tendency of miRNA-miRNA collaboration.
Now, I can’t figure out a way to in vitro validate the collaborative tendency of miRNA of interest with other miRNAs, especially as expression of hundreds of miRNAs in a cell was considered.
