I am using JC 10 stained for mitochondrial membrane potential. if anybody is using can you please suggest me the protocol for staining with JC-10 dye.

I would take a look at what David Nicholls has to say about using JC-1 dye (I'm assuming you mean JC-1, not JC-10).

This dye permeates plasma membranes very slowly, so it does't work well in intact cells. It can be used with good success in isolated mitochondria. Otherwise you might want to choose a different dye, like TMRM for instance.
There are plenty of JC-1 kits out there for measuring mitochondrial membrane potential. Thermo Fisher, Abcam, and Cayman Chemical all have them (with protocols).
