Can anybody suggest to me how to isolate and culture human Hematopoietic stem cells; how to initiate differentiation and what are the parameters required to identify the differentiated cells?
You can found them from peripheral blood. Using flow cytometer you can seperate them with sorting. And then you can easily use them for your experiment.
bone marrow is best, if you can somehow sort them for being CD34 positive (MACS/FACS) thats a good start....

ı am working with HL-60 cell line and ı use ATRA and D3 for diffentiation factor. But they induce HL-60 cells to neutrophil or monocyte like phenotype. which cell type are u working with ?
