I guess you use EM immunostaining to detect NADPH-diaphorase? It should be dot pattern - doesn't it? everywhere in cytosol of neurons? But also you have to define neurons at the ultrastructural level - what criteria do you use for this? Can you send to me some images to have a look.

They are EM preparations. It is a dot pattern labelling after NADPH-diaphorase staining.

Dear Tan, check precipitates in A.C.Cuello Immunohistochemistry Handbook series methods in neurosciences Vol 3. If you did the histochemical reaction with azo-dye methods be aware that your reaction product will be shifted. Look into Pearse histochemistry for EM of azodye techniques. Enrico Marani
