¿Why are both bands present in cells cultured in complete medium recovered after shorter time in culture whereas the upper band seems predominant at longer times? ¿and why the behaviour of the receptor seems to be opposite when cells are cultured in serum free media? Does it correlate with the activation of the receptor and its degradation /endocytosis?
I appreciate any help with it.

Shifted protein bands in WB often indicates posttranslational modification of the protein, for example phosphorylation (search for "phosphorylation-induced mobility shift"). Here I assume that your protein is phosphorylated which is stimulated by the complete media (serum supplemented?) in time-dependent manner.
