In many article ATP level was estimated by Luciferase assay technique but i want other possible method by which we can measure ATP level in cell
Hi I did luciferase assay normalized to protein levels. (Sigma)
Also there are ATP FRET proves tagged to cytosol and mitochondria, they work wuite well!
Hope this helps!
I was using this kit to measure it, simple and easy:

Good luck!
Apart from the enzymatic assay (beetle luciferase), you can use at least the following:
1) NMR (works in live samples)
2) HPLC
3) LC-MS
4) molecular sensors in live cells (based on fluorescent proteins, luciferases, fluorescent dyes)
In all destructive methods beware of the pitfalls (easy ATP degradation during the sample preparation):

Dear Michael
I am extremely thankful to you for your valuable suggestion. can you link paper where ATP were estimated by HPLC or LC-MS. Actually i want to know extraction procedure how it could be estimated by HPLC or LC-MS.
