You can calculate the IC50 by fixing all the other parameters except IC50 in the equation (Hill slope, maximal and minimal % inhibition), but it is not a wise practice. There is important information in the other parameters, especially the Hill slope. Also, there is not enough information to estimate the error in the measurement.
With one fixed concentration IC50 measurement is not possible as we donot know at which concentration 50% cells growth is inhibited. So there is need to check a wide concentration range for IC50 determination.
