Can a drug show cytotoxicity at 24 hours time point but cannot show cytotoxicity at 48 hours in MTT assay? Or there is a possibility that somewhere the experiment has gone wrong? The mode of action of drug is still unknown.
Hello, MTT is a vital cell assay, so there are several possible explanations for the observation of toxicity at 24 but not at 48 hours. For example, if the membranes of the dead cells have burst between 24 and 48 hours then the dead cells will not be visualized and you will see only the viable cells, giving you a false high estimate of viability.
