I am working on the Caco-2 cell line, I'm particularly interested in changes in permeability under inflammatory stimulation.
I am using growth conditions: 12-well transwell plates, seeding density = 70,000 cells pcm^2, growth medium = high glucose DMEM, 10% FBS, strep/penicillin, glutamine.
After 21 days in transwells, each well is displaying TEER values of typically 2000+ ohms. Yet, the literature seems to suggest value of
I had Caco-2s that would be about 200, but found that C2BBe1s were around 50. There is definitely something wrong with your Cacos if they are that high resistance. As these cells are heterogeneous and change after many passages, I would recommend getting new cells from a direct source such as ATCC.

for your answers. It seems that the values I am getting may be down to the passage number (I am using cells from passage 50+) and some dead cells. I will also make sure the chopstick electrodes are cleaned thoroughly before my next measurements.Thank you for alleviating my fears!
