I am planning an experiment to quantify the cellular uptake of peptides in different cell lines by fluorometry. I am referring to a paper from Nature Protocols. They recommended using black 96 well plates. However I don't have them at the moment and would like to perform with normal plates. I did ELISA experiments before with these plates. Please give your opinion on this issue.

I am not sure you if you can use ELISA plates. The crosstalk between wells or interference is too great, and the proteins will stick to you plate because of the coating. I have used regular clear plastic plates and put the samples in every other well. This works. But I would be concerned about the ELISA plates if they will have some special coating. I personally use black coated plates, but white coated work as well.

For fluorometric measurements of enzymatic activities, I used sterile, non-binding 96 black plates. Several maker are available depending on your country. Non-binding plates are the kind of plate you want to avoid any non-wanted interference (I tried myself). However, you can find plates with different types of coating, so you need to check which coating would fit the best to your experimental model.

You may ask for a sample of two or three different maker and make a try.
You have usually 5 to 10 plates in each sterile pack. It is a bit costly, because you have to throw away the plate once used, but it is the best in terms of reliability.
To reduce at maximum the cost, you should plan your experiment in order to use the maximum wells as possible on each plates. (PS: Don't try to use the plate a second time once used, even if washed, it does not work ;).
